چکیده
Abstract Background Polygenic risk scores (PRS) for coronary artery disease (CAD) are emerging as a potential method to improve cardiovascular prediction. Questions remain about their applicability diverse populations well correlation with histopathology. Purpose To assess whether high genetic associates histopathologic plaque morphology. Methods We assessed 122 known CAD loci in 954 Black and White subjects within our sudden death registry generate PRS. The cohort was divided into quintiles according z-score-standardized PRS, both race-stratified fashion the pooled sample. Detailed examination of arteries performed all subjects. Results Subjects highest PRS quintile exhibited more severe atherosclerosis compared lowest quintile, greater mean cross-sectional luminal narrowing (71.5% (95% CI, 66.6%-76.5%) vs. 56.6% 51.1%-62.1%); adjusted p<0.001; Figure 1) higher frequency calcification (adjusted OR 2.19; 95% CI 1.31–3.68; p=0.003) after adjustment first 10 principal components, age, sex, race. Higher predictive finding (i.e., ≥75% narrowing) even additional controlling traditional factors including hypertension, smoking, diabetes mellitus, hyperlipidemia 1.39; 1.19–1.63; 2). Among subjects, associated odds rupture 1.31; 1.03–1.66; p=0.03) predicted CAD-associated cause among younger than 50 years old 1.26; 1.01–1.58; p=0.04). Conclusions This is autopsy study investigating associations between atherosclerotic morphology at level. SUMMARY A polygenic risk score (PRS) is a sum of trait-associated alleles across many genetic loci, typically weighted by effect sizes estimated from a genome-wide association study. The application of PRS has grown in recent years as their utility for detecting shared genetic aetiology among traits has become appreciated; PRS can also be used to establish the presence of a genetic signal in un...
متن کاملMeasuring the effect of risk factors on coronary atherosclerosis: coronary calcium score versus angiographic disease severity.
OBJECTIVES This study sought to determine whether noninvasive quantification of coronary calcium is comparable to selective coronary angiography in measuring the effect of cardiovascular risk factors on coronary atherosclerosis. BACKGROUND Electron beam computed tomography (EBCT) allows the delineation of anatomic coronary atherosclerotic disease and may be useful for noninvasively defining t...

Gross lesions of atheroselerosis in many arterial sites were graded in 500 consecutive autopsies in a general hospital. The grading system used took into account both the total area of intimal involvement and the severity of individual lesions. The first report analyzes the distribution and severity of atherosclerosis in the 347 men and women of the study group who died without morphologic evid...
